In his interview on Club Shay Shay DJ Khaled does go on to confirm that the incident did occur. DJ Khaled goes on to explain why he did not shake Tony Yayo’s hand.

50 Cent dissed Fat Joe on his album The Massacre in his song Piggy Bank.
In addition the 50 Cent beef with Fat Joe had become pretty heated with alleged altercations and disses off track, including Fat Joe’s shot taken at 50 Cent and the 2005 MTV Video Music Awards show.
With all those moving pieces DJ Khaled could not see himself shaking Tony Yayo’s hand. He states how he’s loyal and that shaking Tony Yayo’s hand would be a violation of that loyalty to Fat Joe.
